with a different IC50 value to center on.

5-10-2 Create modified range titration1 layouts

A modified range titration1 is a titration that uses a different stock fluid concentration to expand or
shift the dynamic range of the titration titration. For example, by diluting a 10 mM compound 100X,
the dynamic range available for titration can be shifted by two log units. Another example would be
to use two Dispenseheads, one for the original 10 mM fluid and the other for the diluted 0.1 mM
compound, to allow the dynamic range available for the titration to be extended by two log units,
effectively increasing the dynamic range. Modified range titrations can only be created with the
Wizard if the dispenser is setup to use concentration mode.

If not already in Concentration mode, click Setup/Options.../Units and select the
Concentration radio button as described in

To generate a modified range titration1 with a shifted titration range, follow steps 1 and 2 in
9-1
above, and then click Titration1 in the left pane of the Plate Layout Wizard: Titrations and
Positioning dialog box. This opens the Titration1 pane. click Titration1: <none> in the left pane
of the Plate Layout Wizard: Titrations and Positioning dialog box.

Select the fluid(s) you would like to use for this modified range titration by clicking in the
appropriate Use box for each desired fluid, or click the All button to use all displayed fluids.

For simplicity, set the number of titration zones to 1: click the arrow on the How many titration
zones are there? drop-down list box and select 1 as shown in

Type a value that is below the valid concentration range into the Low (µM) column, for example
0.00002 µM. Note that the number in the table turns red to indicate it is outside the valid range.
