Click the Modified Range button. The Valid Range Modification dialog box appears. See

Click the Fluid Shift option.
Note that a suggestion is made to reduce the fluid concentration to 0.1 mM to enable access to
0.00002 µM concentration with a single drop. Since fluid shift was selected, only one stock
concentration will be used for this fluid, and the maximum concentration is shown as 1 µM due
to the 1% DMSO limit in a 100 uL assay.

Click the Use Modification button, then note that 100 µM High (µM) column entry is now red,
indicating it is outside the new modified range for this fluid. Change the value in the High (µM)
column to the upper limit specified by the Valid Concentration Range display in the dialog box
(1 µM in this case) and the titration is complete. Make sure to remember to dilute the actual
fluid concentration to 0.1 mM when this plate layout is run with the dispenser. The result of this
titration is shown in
Figure
5-28.
