3.6 Precautions on Handling

(e.g. salt solution)

Solvent Displacement and Washing in Flow Path
For displacement in the flow path, mutually soluble solvents
should be used. (Be sure to perform solvent displacement
in the detector cell and sample injector flow path also.)
For washing in the flow path, take the same procedure.
If a rapid replacement is made, salt
precipitation may occur to cause clogging.
Distilled water
Shown above is an example of displacement/washing for
changeover from amino acid analysis to liposoluble vitamin
analysis.

Storage of Column
When leaving the column unused for a long time, fill it with
a solvent and seal its outlet and inlet.
(a) Run a solvent through the column continuously (at a
flow rate of 0.2 mL/min; 0.02 mL/min for a semi-micro
type of column)
(b) Remove the outlet tubing, and attach the sealing plug
to outlet.
(c) Stop the pump.
(d) Remove the inlet tubing, and attach the sealing plug to
the inlet.
(e) Using a wrench, secure the sealing plugs attached at
both ends of the column.
* Before removal, feed an easy-to-elute solvent and

(a) Check the absorption characteristic (wavelength) of a
Polar organic
solvent
(e.g. methanol,
ethanol)
then make displacement with a certain solvent.
