4.2.2 Wavelength accuracy check by using Hg lamp

4.2.3 Sensitivity check

Check the wavelength accuracy by using the bright line at 254
nm of the Hg lamp (see section 3.3.6). "GOOD" will appear on
the display when the wavelength error is within ±3 nm, or "FAIL"
will appear when the error exceeds ±3 nm.
Wavelength calibration is necessary when "FAIL"
NOTE:
appears. See section 4.2.4 (2) for the calibration.
The S/N ratio of the Raman spectrum of water is used to
measure the sensitivity of the fluorescence detector.
The following items must be prepared for the measurement.
The following items are required.
• Pump
• Recorder or D-2500 chromato-data processor
• Distilled water

Injection of Distilled Water
Connect tubing as shown in Fig. 4-1, supply distilled water
at a flow rate of 1 mL/min for 3 minutes with the Chromaster
5110 Pump in Manual mode, then set the flow rate to 0 and
stop the supply of water. Remove the flow cell unit and
check for any bubbles in the flow cell.
If the flow cell has been left empty for awhile (including

when the instrument is installed), first inject methanol
or acetonitrile into the flow cell in order to wet the
interior in advance, then inject distilled water.
Injection of water without first injecting an organic
solvent may cause a reduction of sensitivity.
